Application:

Levocetirizine hydrochloride is a selective histamine H1 receptor antagonist. Mainly used to alleviate allergic symptoms of allergic diseases, clinically used to treat skin and mucosal allergic diseases such as allergic rhinitis, urticaria, angioedema, etc., and also used to alleviate allergic symptoms during colds.

The detailed information is shown in the following English table.

Category Details
Applications

1. Allergic Rhinitis (Seasonal & Perennial):Relieves nasal symptoms (congestion, rhinorrhea, itching, sneezing) and ocular symptoms (pruritus, tearing, redness).

2. Chronic Idiopathic Urticaria:Used to reduce the number, size, and pruritus associated with hives (wheals).

3. Other Allergic Conditions:Off-label use for allergic conjunctivitis and allergic reactions to insect bites.

4. Pediatric Use:offering a safer side effect profile (less sedation) compared to first-generation antihistamines.

5. Veterinary Medicine:Sometimes used to treat atopic dermatitis and pruritus in dogs and cats (off-label or compounded).

KSM Synthetic Intermediates

1. (R)-2-Chloropheniramine (or related chiral amine):Used in asymmetric synthesis routes to introduce the chiral center early in the process.

2. 4-Chlorobenzhydryl Chloride (CAS 134-83-8):A common intermediate in antihistamine synthesis; reacts with ethanolamine derivatives to form the diphenylmethyl ether linkage.

3. 2-[2-[4-[(4-Chlorophenyl)phenylmethyl]-1-piperazinyl]ethoxy]acetic acid (Racemic Cetirizine, CAS 83881-51-0):The racemic mixture; used as the starting material in resolution processes where Levocetirizine is separated using chiral acids (e.g., dibenzoyl-D-tartaric acid).

4. (R)-Cetirizine Base (CAS 100643-71-8):The free base form of Levocetirizine, which is then converted to the dihydrochloride salt for stability and solubility.

5. Chiral Resolving Agents (e.g., Dibenzoyl-D-tartaric acid, CAS 17026-42-5):Crucial for the resolution of racemic cetirizine to isolate the active (R)-isomer.
