Product Introduction & Application:
Colestyramine, also known as cholesterol reducing resin, cholesteryl amine reducing resin, and lipid-lowering resin 1, is a strong alkaline quaternary amine anion exchange resin used in low-density lipoprotein elevation hyperlipidemia, namely type IIa and IIb hyperlipidemia, and is the most effective cholesterol lowering drug. After oral administration, it is not absorbed and exchanges chloride ions with bile acids in the intestinal lumen, stably binding with bile acids until it is excreted with feces. This reduces the absorption of exogenous cholesterol, hinders the hepatic intestinal circulation of bile acids, and reduces the amount of bile acids in the blood. As a result, it promotes the conversion of cholesterol to bile acids in the blood, thereby reducing cholesterol. Colestyramine has no effect on reducing triglycerides. Due to its powerful cholesterol lowering effect, this product has become the preferred medication for type IIa hyperlipidemia. For type IIb hyperlipidemia, it should be combined with drugs that can reduce triglycerides in the blood, such as clopidogrel or niacin. For type III, IV, and V hyperlipidemia, it is not advisable to use cholestyramine. In addition, this product can also be used to treat skin itching caused by atherosclerosis, cirrhosis and cholelithiasis.
Intermediates of Colestyramine:
| Product Name | CAS No. | Role in Colestyramine Synthesis |
|---|---|---|
| Styrene | 100-42-5 | Main backbone monomer: Polymerizes to form the polystyrene matrix. |
| Divinylbenzene (DVB) | 1321-74-0 | Cross-linking agent: Cross-links polystyrene chains to form an insoluble 3D resin network. |
| Chloromethylstyrene (CMS) | 30030-16-7 | Functional intermediate: Reacts with styrene/DVB to introduce –CH₂Cl groups onto the polymer backbone. |
| Trimethylamine (TMA) | 75-50-3 | Quaternizing agent: Aminates –CH₂Cl groups to form benzyltrimethylammonium chloride (quaternary ammonium active sites). |
| Dimethyl sulfate (DMS) | 77-78-1 | Alternative quaternizing agent: Methylates amine groups to form quaternary ammonium salts. |
| Styrene–divinylbenzene copolymer | - | Polymer intermediate: Cross-linked porous resin before quaternization. |
| Chloromethylated polystyrene resin | - | Styrene–DVB copolymer with reactive –CH₂Cl groups. |
| Hydrochloric acid (HCl) | 7647-01-0 | Provides Cl⁻ counterion for quaternary ammonium groups. |
